大赖草DNA导入小麦获得大穗株系育性及其细胞学初步研究

摘    要:对大赖草DNA导入小麦后代中选出的部分大穗材料,进行了育性和花粉母细胞(PMC)减数分裂的细胞学鉴定.结果表明:人工套袋自交不育率平均高达93.01;,天然异交不育率为36.4;~100;;减数分裂后期出现了桥、环及落伍染色体,这些异常现象说明了转化后代的染色体结构发生了变化,大赖草DNA片段已整合到染色体上;DNA片段的导入产生单价染色体,这是遗传学上的新问题,其原因尚需进一步研究确定.

Abstract:The fertility of big spike wheat lines elected in the generation with transgene of Leymus racemosus(Lam.)Tzvel was studied,and pollen mother cell was identified in meiosis.The result showed that the average sterile rate of self-crossing sterility was up to 93.01% by hand bagging,sterile rate of nature-crossing was from 36.4% to 100%.The bridge,ring and behindhand chromosome appeared later meiosis stage.The abnormal phenomena explained the chromosome structure of transformation generation changed.The genetic fragment had already combined on the wheat chromosome.The insertion of the genetic fragment produced the univalent chromosome.It is a new problem in science of genetics,its reason still needs to determine further.
