玉米矮花叶病毒HC-Pro在蚜虫传毒过程中的作用机制

摘    要: 本文用亲合印迹法测定了玉米矮花叶病毒(MDMV)与MDMVHC-Pro,及二者与蚜虫蛋白之间的互作。结果表明,MDMV HC-Pro可以与MDMV结合,也可以直接与MDMV的介体——麦二叉蚜(Schizaphis graminum)中分子量约为56kD的可溶性蛋白结合,而MDMV不能与麦二叉蚜的可溶性蛋白直接结合。无论是MDMV HC-Pro,还是MDMV,都不能结合非介体灰飞虱(Laodelphax striatellus)的可溶性蛋白。酶联板模型的结果与此一致。上述结果说明,病毒不能直接与蚜虫口针中的病毒附着位点(VAS)结合;HC-Pro一端连接病毒,一端连接VAS,在蚜虫传播MDMV的过程中起桥梁作用。这是关于病毒HC-Pro与介体蚜虫蛋白直接互作的第一个生化证据。

Abstract:The interactions between maize dwarf mosaic virus (MDMV), HC Pro, and virus attachment site (VAS) were studied by affinity blot (overlay assay of virus protein blot). MDMV HC Pro could bind MDMV in the assay, and vise versa. Only MDMV HC Pro could react with the soluble protein of aphid vectors directly, while MDMV was not. It therefore provided evidence to support the hypothesis that HC Pro acted as a bridge in aphid transmission of virus. Neither CP nor HC Pro was able to interact with any protein of non vectors. The mole cular weight of the aphid protein interacting with MDMV HC Pro was ca. 56 kD. This is the first biochemical evidence of interaction between HC Pro and VAS in aphids.
