越橘果实总RNA提取方法比较研究

摘    要:越橘成熟果实富含多糖、多酚和花色素苷等次生代谢物质,严重影响果实总RNA的提取.试验以越橘成熟果实的果肉和果皮为试材,采用适合多糖多酚植物RNA提取试剂盒及试剂和改良的CTAB法提取越橘果肉和果皮中总RNA,经琼脂糖凝胶电泳和核酸蛋白测定仪(NanoDrop 2000)检测.结果表明,改良的CTAB法效果最好,提取的总RNA完整性好,纯度高.通过RT-PCR验证,所提取的总RNA可以满足基因克隆等分子生物学试验的要求.

Abstract:It is difficult to isolate total RNA from blueberry fruit because of a great deal of polysaccharides,polyphenolics,anthocyanins and other metabolites.In this study,total RNA from pulp and skin of blueberry fruit was isolated with plant RNA Isolating Reagent kit,reagent method and improved CTAB method.The total RNA were detected by the agarose gel electrophoresis and nucleic acid/protein assay(NanoDrop 2000).The results showed that improved CTAB method had the best result.The total RNA isolated by this method was proved to be relatively intact and with high purity.The isolated RNA could be used in molecular biology experimentation by RT-PCR analysis.
