三个花生二酰甘油酰基转移酶基因的克隆与序列分析

摘    要:二酰甘油酰基转移酶在植物油脂合成途径中具有重要的作用。本研究通过构建花生种子全长cDNA文库,结合大规模EST测序和RACE克隆等方法,从花生中克隆得到了三个DGAT基因,分别命名为AhDGAT1-1、AhDGAT1-2和AhDGAT3-3。其中AhDGAT1-1全长为2020bp,ORF为1539bp,理论分子量为58.6036kD,理论等电点为8.83;AhDGAT1-2全长为2553bp,ORF为1581bp,理论分子量为60.6598kD,理论等电点为8.94;AhDGAT3-3全长为1377bp,ORF为1023bp,理论分子量为36.911kD,理论等电点为8.17。将这三个基因在GenBank上注册,注册号分别为KC736068,KC736069和KC736067。

Abstract:Diacylglycerol acyltransferase (DGAT) is a key enzyme in biosynthesis pathway of veg-etable oil in plant.We constructed a full-length cDNA library of peanut (Arachis hypogaea L.) seed via a large number of sequences of expressed sequence tag (EST) and RACE clone, three diacylglycer-ol acyltransferase genes, designated AhDGAT 1-1, AhDGAT 1-2 and AhDGAT 3-3 were isolated from peanut.The sequence of AhDGAT 1-1 cDNA was 2020 bp, and ORF was 1539 bp.The molecular weight of AhDGAT1-1 was 58.6036 kD and its isoelectric point (pI) was 8.83.The sequence of Ah-DGAT 1-2 and AhDGAT 3-3 cDNA were 2553 bp and 1377 bp, and ORF were 1581 bp and 1 1023 bp, respectively.The molecular weight of AhDGAT1-2 and AhDGAT3-3 were 60.6598 kD and 36.91 1 kD, and isoelectric points (pI) were 8.94 and 8.17, respecively.The sequences of the three genes were also submitted to GenBank and their GenBank ID were KC736068, KC736069, KC736067, re-spectively.
