摘    要:分别用冻干、常温干燥器干燥、36℃烘干、液氮冷冻、-80℃冷冻等5种方法对海南高种椰子花粉的电子扫描显微镜观察样品进行预处理。结果表明:以冻干法的效果最好;常温干燥器干燥与36℃烘干法制备的花粉样品在扫描电镜下观察呈现塌陷、变形状;-80℃冷冻与液氮冷冻处理的雄花外壁呈褐色,花药、花丝潮湿,电镜观察视野中的杂质、碎片较多。

Abstract:Five methods(freeze-drying,silica-gel drying at room-temperature,36 ℃ drying,liquid nitrogen freezing,-80 ℃ freezing) were used to pretreat the male flowers of coconut,and then scanning electron microscope(SEM) was used to observe the morphology of the pollen.The results showed that the freeze-drying pretreatment had the best effect for the observation of coconut pollen,and the exine ornamentations of the pollens pretreated by freeze-drying were clear.While some of the pollens pretreated by 36 ℃ drying or s...
