不同物种Dock7基因编码区生物信息学分析

摘    要:为了研究不同物种Dock7基因的遗传多样性,采用生物信息学的方法分析了人、倭黑猩猩、大猩猩、毛猩猩、东非狒狒、玻利维亚的灰鼠猴子、虎鲸、家马、白犀牛、雪貂、家牛、大熊猫、佛罗里达海牛、海象、欧洲兔、家猫、家绵羊、星鼻鼹鼠、北美鼠兔、裸鼢鼠、非洲跳鼠、八齿鼠、褐家鼠、小家鼠、袋獾、原鸡、爪蟾27个物种Dock7基因编码区(CDS),并对该基因的遗传多样性、氨基酸序列、信号肽、跨膜结构域、疏水性/亲水性、蛋白质二级结构进行了分析和预测。结果表明,在27个物种68条基因序列中共检测到1855个多态位点,生成28种单倍型,Dock7基因序列编码区种间存在丰富的遗传多样性,而种内变异极小,只在原鸡中发现一个变异位点。在所研究27个物种中,原鸡Dock7基因的氨基酸序列存在信号肽,其他26个物种Dock7基因的氨基酸序列均无信号肽。27个物种都没有跨膜结构域,表现为亲水性,蛋白质二级结构主要结构元件是自由卷曲和α-螺旋,Dock7蛋白呈酸性。

Abstract:To study the genetic diversity of Dock7 gene in different species, the CDS of Dock7 gene sequences from Homo sapiens, Pan paniscus, Gorilla gorilla gorilla, Pongo abelii, Papio anubis, Saimiri boliviensis boliviensis, Orcinus orca, Equus caballus, Ceratotherium simum simum, Mustela putorius furo, Bos taurus, Ailuropoda melanoleuca, Trichechus manatus latirostris, Odobenus rosmarus divergens, Oryctolagus cuniculus, Felis catus, Ovis aries, Condylura cristata,Ochotona princeps, Heterocephalus glaber,Jaculus jaculus,Octodon degus, Rattus norvegicus, Mus musculus, Sarcophilus harrisii, Gallus gallus, Xenopus tropicalis were analyzed using the method of bioinforma- tics. The genetic diversity, amino acid sequences, signal peptide, transmembrane topological structure, hydro- hobieity or hydrophilicity and protein secondary structure were also analyzed. The results showed that a total of 1855 polymorphie sites were detected from 68 sequences of 27 species, from which 28 hapolotypes were sorted, the genetic diversity of the Dock7 gene CDS was rich within and among species. In the 27 species, the amino acid sequences of Dock7 gene of the Gallus gallus have signal peptide but others haven't. The Dock7 protein was a no transmembrane protein and the polypeptide chain presented hydrophilic, the main motifs of predicted secondary structure of Dock7 are random coil and alpha helix, Dock7 protein is acidic.
