稻瘟病菌磷酸甘露糖基转移酶(PMT)蛋白家族 基因结构特征分析

摘    要:为阐述稻瘟病菌(Magnaporthe oryzae)磷酸甘露糖基转移酶PMT基因的结构特征及可能的生物学功能,采用序列比对分析方法,明确了稻瘟病菌PMT蛋白家族成员在氨基酸水平的保守性。研究发现它们与来自灰葡萄孢(Botrytis cinerea)、粗糙脉孢霉菌(Neurospora crassa)、玉米赤霉(Gibberella zeae)及构巢曲霉(Aspergillus nidulans)等丝状真菌的PMT蛋白高度同源;进一步生物信息学分析表明,稻瘟病菌PMT蛋白不含有信号肽,为非分泌蛋白,含有跨膜结构域,作用于内质网膜上,具有保守的PMT motif 和MIR motif。研究结果可为进一步阐明PMT蛋白在稻瘟病菌生长发育及致病过程中的作用机理奠定基础。

Abstract:To clarify the structure characteristics and the possible biological functions of the dolichyl- phosphate-mannose-protein mannosyltransferase encoding gene PMTs in Magnaporthe oryzae, the sequences of PMTs were analyzed. The results showed that the MoPMTs are conservative in the structure of amino acid and highly homologous with the PMT protein from Botrytis cinerea, Neurospora crassa, Gibberella zeae, and Aspergillus nidulans. Further bioinformatics analysis indicated the protein of PMT is a nonsecreted protein without a signal peptide, contains transmembrane domains, and localices at the endoplasmic reticulum, and has conserved domain of PMT motif and MIR motif. These results will facilitate the further studies on the mechanism of PMTs in M. oryzae.
