金丝小枣组织培养快速繁殖的研究

摘    要:该文通过对良种金丝小枣组织培养快速繁殖的研究,选出了枣树组培的最佳外植体,并首次将WP培养基应用于枣树组培,筛选出最适的启动、增殖和生根培养基,使其试管苗的增殖在40d内达到4.5倍,生根率达到90%以上。在生根苗移栽试验中,摸索出了适合的基质及移栽管理程序,移栽成活率达96%,试验还就光照方向、温度及继代周期对枣树组培的影响进行了探索。

Abstract:Fast propagation of Chinese date ‘Jinsi Xiaozao’ with tissue culture is studied.The buds from the cuttings of one or two-year-old root sprouts that forced germination inwater culture are the optimal explants;Lioydant and McCowns’(1981)Wood Plant(WP)medium is used in jujube tissue culture for the first time,the optimal media of initial, prolif-erating and rooting inducing cuIture are found,the proliferating rate increases 4.5 times af-ter 40 days subculture than subcuIture beginning,the rooting rate is over 90%in root induc-tion culture. Suitable transplanting substrate and transplanting procedure are explored intransplanting plantlet experiment and transplanting survival rate of plantlets gets 96%,Inaddition,effect of illuminating direction, temperature and subculturing frequency on thegrowth of shoots is also studied.
