鸭PRLR基因拷贝数的变异

摘    要:本研究以台湾褐色菜鸭为试验动物,采用荧光定量PCR技术,使用2-ΔΔCt换算法对PRLR基因拷贝数进行定量,分析了其多态性与台湾褐色菜鸭生产性能的相关性.结果表明,PRLR和Ldh-B标准曲线的R2分别为0.991和0.990,扩增效率分别为111.68%和108.429%,斜率分别为-3.070和-3.135,PRLR和Ldh-B的扩增效率近似一致,可通过2-ΔΔCt方法对PRLR基因进行定量分析.在94羽台湾褐色菜鸭的样品中共检测到5种拷贝数(1、2、3、4、5)变异类型,拷贝数变异与台湾褐色菜鸭的蛋壳厚度和蛋形指数显著相关(P0.05),拷贝数为2的个体蛋壳厚度显著高于拷贝数为1和5的个体;拷贝数为3的个体蛋形指数显著低于拷贝数为1、2和4的个体;其余性状均无显著相关(P0.05).因此,PRLR基因拷贝变异区域可能影响蛋壳厚度和蛋形指数.

Variation of PRLR gene copy number in duck
Abstract:Copy number variation ( CNV) merges to be an effective tool in poultry breeding. To elucidate the correlation between productive performance and polymorphism, gene of Brown Tsaiya ducks were amplified by fluorescence quantitative PCR, and fol-lowed by being transferred into CNV via 2-ΔΔCt method. Subsequent production traits responses to different CNV of PRLR gene were investigated. Results showed that correlation coefficients of standard curve for PRLR and Ldh-B were 0.991 and 0.990, with slopes of standard curves being-3.070 and-3.135, respectively. Amplification efficiency of PRLR and Ldh-B target gene were 111.68% and 108.429%, which were approximately at the same level with reference gene. Traits of Brown Tsaiya ducks were correlated with CNV ( P<0.05) , basing on which CNV were classified into 5 levels, named copy number 1 ( CN 1) , CN 2 and so forth. Association anal-ysis of early production traits showed that shell thickness of individuals with CN 2 were significantly higher than that of CN 1 and CN 5 ( P<0.05) and egg shape index of individuals from CN 3 were significantly less than that of individuals with CN 1, 2 and 4 copy number for ( P<0.05) . Therefore, copy number variable regions in PRLR gene might affect eggshell thickness and egg-shape index.
