摘 要: | 以温室种植的‘秋香’番茄健壮幼嫩侧芽为材料,研究4种不同消毒方法对番茄外植体的消毒效果,并通过继代培养,进一步研究并筛选‘秋香’番茄组织培养最佳培养基。试验结果表明:外植体消毒效果较理想的方式是10%的NaClO溶液(v/v)消毒20min,后用0.1%HgCl_2溶液(w/v)+2滴吐温20消毒3min;无菌株系建立适合的培养基为MS+BA 0.1mg·L~(-1)+IBA 0.1mg·L~(-1);继代培养适宜培养基配方为MS+BA0.1mg·L~(-1)+IBA 0.5mg·L~(-1),可以较快获得健壮的‘秋香’番茄试管苗。

Abstract: | Four methods for disinfecting the tomato explants were first compared on their efficacies prior toapplying the sanitized tender lateral buds from Qiuxiang tomatoes (Lycopersicon esculentum var.cerasiforme ) in the greenhouse for the propagation experiment.The selected disinfection treated the explants in a 10% NaClO solutionfor 20 min,followed by another of 0.1% mercuric chloride with a small amount of Tween 20 for 3 min. Theoptimized culture mediumfor establishing the aseptic lines consisted of MS+BA 0.1 mg·L-1 +IBA 0.1 ·L-1 . Whereas,the medium made of MS+BA 0.1 ·L-1 +IBA 0.5 ·L-1 was found to be suitable for the subculture toyield newhealthy and strong Qiuxiang tomato plantlets. |
