摘    要:【目的】从分子水平上了解带叶兜兰的遗传多样性,为带叶兜兰种质资源的保护和利用奠定基础。【方法】采用SRAP分子标记技术,对40份采自广西木论自然保护区天然居群的野生带叶兜兰种质资源进行分析,利用Popgene软件估算遗传多样性参数。【结果】从414对SRAP引物中筛选获得30对能扩增稳定、清晰可辨条带的引物组合;30对引物组合可从40份DNA样品中扩增获得256条带,每对引物扩增条带数为6~11条,平均每对引物组合扩增获得8.53条带;其中多态性条带62条,多态性比例为12.50%~33.33%,平均多态性比例为24.22%。遗传多样性分析结果显示,Nei’s基因多样性指数为0.1282,Shannon多样性指数为0.1582。【结论】带叶兜兰种质资源群体个体数少,个体间遗传多样性水平较低。

Abstract:【Objective 】The genetic diversity of Paphiopedilum hirsutissimum was analyzed on the molecular level in order to lay the foundation for protection and application of its germplasm resources. 【Method 】The genetic diversities of40 wild germplasm resources of Paphiopedilum hirsutissimum collected from Guangxi Mulun natural reserve area were analyzed by using SRAP marker technology. The indices of genetic diversity were estimated by POPGENE software.【Results 】Among 414 pairs of SRAP-PCR primers, 30 pairs of primer combinations could amplify stable and clear discernible bands. By using 30 pairs of primer combinations, 256 bands could be amplified from 40 DNA samples.Each pair primer could amplify 6-11 bands and the average was 8.53 bands. Out of 256 bands, 62 bands were polymorphic, which accounted for 12.50%-33.33% with an average of 24.22%. The results of genetic diversity indi-cated that Nei's gene diversity index was 0.1282, and Shannon's information index was 0.1582. 【Conclusion】There is low level of geneticst diversity amongst individuals of Paphiopedilum hirsutissimum germplasm resources being less population individuals.
