| 摘 要： | 从猪十二指肠肠道组织提取总RNA，采用RT-PCR方法克隆了猪Sirtuin3基因，并与GenBank中的9个其他物种的核酸序列进行了同源性分析，采用邻接法构建了分子系统进化树。结果表明克隆的猪Sirtuin3基因长999bp，编码332个氨基酸，前21个为信号肽；猪Sirtuin3与牛、人的同源性最高，分别为86．3％、79．6％，分子进化树也表明猪的Sirtuin3与牛、人的亲缘关系最近。

| Abstract: | Total RNA was isolated from porcine duodenal perforation tissue.The Sirtuin3 cDNA was amplified by RT-PCR and then cloned and sequenced.The nucleotide sequence was compared with the homologous genes in other species retrieved from the GenBank,and finally the phylogenetic tree was constructed by NJ method.The results indicated that the cloned porcine Sirtuin 3 gene was 999 bp in length,the deduced amino acids had 332,and the signal peptide of predicted amino acid sequence was the 1~21st amino acid.Homology analysis showed that the sequence shared 86.3% and 79.6% homology with those of cow and human respectively.Phylogenetic analysis based on the nucleotide sequence from the Sirtuin3 gene showed that pig had a closer phylogenetic relation with cow and human. |
