芦荟原生质体的分离

目的]为进一步的离体培养快速繁殖奠定基础,同时也为原生质体的融合(体细胞杂交)提供条件。方法]采用酶解的方法以芦荟的叶片为材料进行原生质体分离。结果]用芦荟的叶肉细胞可成功分离出原生质体,而且活力较高,可达90%以上。结论]芦荟原生质体的成功分离主要取决于材料的来源、酶液的组成。另外还受酶解液的渗透压、温度和pH值影响。

Protoplast Isolation of Aloe vera L.

Objective] The purpose was to lay a foundation for repaid propagation of further culture in vitro,and provide conditions for protoplast fusion(somatic hybridization) of Aloe vera L.as well.Method]With the leaf A.vera as material,the protoplast was isolated by using enzymatic hydrolysis.Result] The protoplast could be isolated successfully with mesophyll cell of Aloe,and activity of protoplast was high,which was over 90%.Conclusion] The successful protoplast isolation of A.vera mainly depended on the material sources,composition of enzyme solution.It was also affected by osmotic pressure of enzyme solution,temperature and pH value.
