南方红豆杉人工林药材活性成分指纹图谱鉴别研究

摘    要:利用高效液相色谱(HPLC)指纹图谱技术,建立HPLC指纹图谱的色谱条件,研究了重庆地区11批6年生栽培南方红豆杉枝叶色谱指纹图谱的共有特征规律。结果表明:SHIMADZU C18色谱柱(250mm×4.6μm,ID5μm),乙腈和水为流动相进行梯度洗脱,波长227nm,流速0.8mL/min为HPLC指纹图谱的最佳色谱条件;在此色谱条件下,栽培南方红豆杉色谱指纹图谱中共有17个共有色谱峰,其中峰9可作为参照峰;共有峰相对保留时间和相对峰面积的平均RSD均小于2.0%;非共有峰面积在4.1%~8.3%之间,符合色谱指纹图谱研究规定小于10%的检测要求;全图谱共有峰的相似度均≥93%。表明所建立指纹鉴别方法具有稳定性高、精度准、重现性好,选定的HPLC指纹图谱可以对栽培南方红豆杉药材组分的一致性作出可靠的判断。

Abstract:A HPLC chromatographic fingerprint system for the cultivated Taxus chinensis var. mairei was established based on the chromatographic fingerprint characteristics of leaves and branches from 11 batches of the 6-year cultivars in the Chongqing region. The optimum chromatography conditions are as follows: a SHIMADZU C18 column (250 mm×4.6 μm, 5 μm), a mobile phase of acetonitrile and water as gradient elution, a detective wave-length of 227 nm and a flow rate of 0.8 mL/min. Under these conditions, a total of 17 shared peaks were detected, of which No.9 was selected as the reference peak. The average relative standard deviation (RSD) of both the relative retention time and the relative area of shared peaks was lower than 2.0%. The unshared peak area occupied 4.1%--8.3% of the total, which conforms to the requirement of chromatographic fingerprint regulations (lower than 10%). Correlation analysis showed that the similarity of the shared peak was ≥93%, indicating that this chromatographic fingerprint system is stable, accurate and reproducible and can be recommended for identification of cultivated T. chinensis var. mairei.
