野生泸定百合鳞片的组织培养技术

摘    要:为野生泸定百合资源的保护及开发应用提供依据,以其鳞片为外植体,观察其不定芽的诱导、增殖以及生根培养情况。结果表明:野生泸定百合鳞片用0.1%升汞消毒15 min,效果较好;MS+6-BA1.0mg/L+NAA0.1mg/L为野生泸定百合鳞片最佳不定芽诱导培养基,平均每鳞片分化芽数达5.4个;MS+6-BA 2.0mg/L+NAA 0.05mg/L为鳞片最佳增殖培养基,增殖系数达7.66;1/2MS+IBA 1.0mg/L为鳞片最佳生根培养基,生根率达95.2%,平均根数5.3条,平均根长3.9cm。

Abstract:In order to provide references for protection,development and utilization of wild L.sargentliae resources,with bulb scales of wild L.sargentliae as explants,the effects of the hormones on the adventitious bud induction,proliferation and rooting were observed.Results:The best disinfection method of bulb scales of wild L.sargentliae was using 0.1% mercuric chloride for 15 min.MS+6-BA 1.0 mg/L+NAA 0.1 mg/L medium was the best adventitious bulbs induction medium,the average number of buds per scale was 5.4.MS+6-BA 2.0 mg/L+NAA 0.05 mg/L was the best proliferation medium,and increment coefficient was up to 7.66.1/2 MS+IBA 1.0 mg/L was the best rooting medium,the rooting rate was 95.2%,the average root number was 5.3,and the average root length was 3.9 cm.
