聚乙二醇对七叶一枝花种子快速萌发效果的影响

摘    要:为探明聚乙二醇(PEG)不同分子量及体积分数对七叶一枝花种子萌发的效果,采用不同体积分数(10%、25%、35%)和分子量(4 000、6 000、8 000、10 000)的聚乙二醇浸泡七叶一枝花种子,以蒸馏水浸泡作对照(CK),放入20℃培养箱(弱光条件)24h后,清洗种子后进行培养,并研究去掉种子部分种皮、胚乳对子叶及切去种子茎对侧芽的影响。结果表明:PEG对七叶一枝花种子的快速萌发具有促进作用,其中以25%PEG 4000促进七叶一枝花当年采集新种子的萌发效果最好,培养30d、40d、50d和60d时萌发率分别为12.61%、44.85%、64.38%和78.86%,比CK高2.88百分点、29.69百分点、17.97百分点和5.33百分点;70d时萌发率达85.71%,最长胚根长达1cm;80d时萌发率达90%以上,最长胚根达3cm。为达到较好的药用效果,建议在培养期间去掉种子部分种皮和切去生长的茎。

Abstract:P.polyphylla seeds soaked with different molecular weight and volume fraction of PEG were cultured in an incubator at 20℃ for 24 h to discuss the effect of PEG on germination of P.polyphylla seeds.New P.polyphylla seeds were used to study the effect of removed partial seed shell or endosperm on cotyledon growth and the effect of removed differentiation stem on growth of lateral buds.Results: PEG has the promotion effect on rapid germination of P.polyphylla seeds.25% PEG with 4 000 molecular weight treatment has the best promotion effect on germination of new P.polyphylla seeds among all treatments and the germination rate of new P.polyphylla seeds soaked with 25% PEG with 4 000 molecular weight after 30 d, 40 d 50 d and 60 d is 12.61%,44.85%, 64.38% and 78.86%, 2.88%, 29.69%, 17.97% and 5.33% higher than CK (distilled water treatment) respectively.The germination rate and the longest radicle length of new P.polyphylla seeds soaked with 25% PEG with 4 000 molecular weight after 70 d and 80 d are 85.71% and 1cm, and above 90% and 3cm respectively.To remove partial seed shell and cut differentiation stem during the culture period of P.polyphylla seeds can reach the better medicinal effect.
