毛果杨Eukaryotic translation initiation factor 5A(eIF5A) 同源基因的生物信息学分析

摘    要:为研究毛果杨eIF5A基因间的同源性及其进化关系,以毛果杨的eIF5A基因为研究对象,利用生物信息学软件及网站对其进行碱基分布、氨基酸组成、亲疏水性、保守区以及二级结构和三级结构的预测与分析,并与其他物种的eIF5A氨基酸序列进行多重比对与进化分析。结果表明,4个毛果杨eIF5A基因定位于不同染色体上,且都只含有5个外显子;研究还发现不同成员间氨基酸数目、氨基酸序列间的疏水性存在一定的差异;亚细胞定位分析表明,4个eIF5A蛋白均定位于细胞质上;二级结构预测结果显示,4个eIF5A氨基酸序列以无规则卷曲、扩展链和α-螺旋为主要组成部分,且4条氨基酸序列三维结构十分相似。上述结果均为毛果杨eIF5A基因家族的进一步功能分析提供了一定基础。

Abstract:To study the homology and evolutionary relationship among eIF5A gene of Populus trichocarpa, bioinformatics softwares and websites were used to predict and analyze its base distribution, amino acid composition, hydrophilcity or hydrophobicity, secondary structure and tertiary structure. A polygenetic tree was constructed based on the corresponding eIF5A proteins from other species, and the conserved sequences in these proteins were analyzed. The results showed that four eIF5A genes were located on different chromosomes and all had five exons. The research still found that the amino acid number and sequence hydrophobic in eIF5A gene family presented some differences. Protein subcellular localization analysis showed that all members of eIF5A were located in the cytoplasm. Furthermore, the main composition of 4 amino acid sequences were random coil, extended strand and alpha helix via secondary structure prediction, and they were similar to each other in three-level structure. The results of this investigation could definitely provide some foundation for further function analysis of eIF5A gene family.
