鹅源副黏病毒HN基因真核表达质粒的构建及基因疫苗的研究

摘    要:本试验通过FCR技术从pGEM-HN质粒中扩增出了HR基因,构建了真核表达质粒pcDNA3-HN。真核表达质粒pcDNA3-HN肌肉注射免疫鸡后,HI血清抗体效价较空白对照组高,攻毒后鸡发病、死亡均少于空白对照组,有27%-36%的鸡耐过。但血清抗体效价较灭活苗组低,发病率和死亡率较灭活苗组高。试验结果表明HN基因在鸡体内,得到了表达,并使鸡获得了一定的免疫力,但保护作用不及鹅副黏病毒油乳剂灭活苗。

Abstract:In this report, we amplified HN gene by PCR from the plasmid pGEM-HN, and constructed a eukaryotic expression plasmid pcDNA3-HN. The DNA vaccine was administrated by intramuscular injection to chickens. The HI serum antibodies titer was obviously higher than control. Morbility and fatality were less than control after challenged by GPMV, and 27~36 per cent chickens survived. However, the HI serum antibodies titer was lower than inactive vaccine group and morbility and fatality were more than it. It is shown that HN gene was expressed in chicken and induced protective immunity, but it's protection is less than inactive vaccine of GPMV.
