| 摘 要： | 建立猪戊型肝炎病毒(Swine Hepatitis E Virus,SHEV)RT-PCR检测方法,为SHEV的流行病学调查、临床诊断提供可靠的技术手段。根据SHEV的基因组序列,设计并合成2对引物,以SHEV江西分离株SHEV-JX-1为模板,对反应条件进行了优化,建立了诊断SHEV的RT-PCR方法。该方法可从SHEV-JX-1中扩增出230 bp的片段,测序结果表明为SHEVⅣ型基因片段;对猪的其它病毒进行检测结果均为阴性;能检测出2.5×10-8μg的SHEV RNA;对江西部分猪场采集的胆汁及肠容物进行检测,结果胆汁中的阳性率为10%,肠容物的阳性率为4%。实验建立的RT-PCR方法具有敏感、特异、重复性好的特点,可用于SHEV的分子流行病学调查和临床快速诊断。
