摘    要:以普通果球和少果球悬铃木为供试材料,建立了叶片不定芽再生体系。结果表明:种子苗于附加IBA0.01 mg/L、ZT 0.10 mg/L及6BA 0.10 mg/L的WPM基础培养基上扩繁,叶片小而多,叶色深绿;取扩繁苗倒1~5叶片外植体,于附加不同浓度6BA、IBA及KT的WPM培养基上,普通果球和少果球悬铃木最高不定芽再生频率分别达59.5%和100.0%,每个外植体长芽数分别近5个和2个。再生所需最佳激素组合因品种而异,普通果球和少果球悬铃木分别为IBA 0.2 mg/L、6BA 2.0 mg/L及KT 0.5 mg/L和IBA 0.5 mg/L、6BA 2.0 mg/L及KT 0.5 mg/L;不定芽及芽点在WPM 0.01 mg/L IBA 0.10 mg/L ZT 0.10 mg/L 6BA伸长培养基上迅速伸长,且株体健壮;在WPM 0.2 mg/L IBA 0.5%活性炭生根培养基上,根系和上部发育较好。

Abstract:Adventitious shoots were successfully regenerated from leaf explants of in vitro cultures of Platanus orientalis Willd.varieties with common fruits and fewer fruits separately.Explants of five uppermost expanding leaves of two genotypes were cultured on four media based on WPM basal medium containing different concentrations of 6BA,IBA and KT.The highest regeneration rates of shoot clumps per leaf explant were 59.5% and 100.0% for Platanus orientalis Willd.varieties with common fruits and fewer fruits,and the largest number of shoot clumps were five and two,respectively.Best regulator combinations for regeneration of adventitious shoot were IBA(0.2 mg/L),6BA(2.0 mg/L) and KT(0.5 mg/L),and IBA(0.5 mg/L),6BA(2.0 mg/L) and KT(0.5 mg/L) for Platanus orientalis Willd.varieties with common fruits and fewer fruits.Adventitious shoot grew rapidly in WPM medium added with(0.01 mg/L) IBA,(0.1 mg/L) ZT and(0.1 mg/L) 6BA.The regenerated shoots were micropropagated and rooted successfully.
