摘    要:用3种不同方法提取辣椒叶片的总RNA,研究辣椒叶龄和基因型对总RNA提取的影响和筛选辣椒总RNA提取的最佳方法.结果表明,方法2为辣椒叶片总RNA提取的最佳方法;从新叶中提取的总RNA的亮度高,完整性最好,中龄叶次之,老叶最差;辣椒材料0860-1总RNA的28 S和18 S条带明亮,质量浓度最高,材料0876和08122的这些条带较暗,质量浓度低.

Abstract:Three methods were applied to extract total RNA from the leaves in order to understand the effects of leaf ages and genotypes and choose the best method for total RNA extraction in hot pepper in the present study. The results showed that the 28 S and 18 S bands extracted by method 2 were strong, and the quality and concentration of the total RNA were high. The lightness and quality of the total RNA from young leaves extracted by method 2 were the highest, next came medium leaves, and then old ones. The 28 S and 18 S bands from the leaves of line 0860-1 extracted by method 2 were bright, and total RNA concentration was the highest; but those of the lines 0876 and 08122 were dark, and the concentration was low. Therefore, the extraction effect of total RNA might be influenced by hot pepper genotypes.
