枇杷胚乳离体培养获得植株的研究

摘    要:在观察、研究枇杷(Eriobotrya japonica Lindl.)幼果的外部形态特征及果皮颜色的变化与其种子内胚胎发育进程相关性的基础上,根据幼果外观特征采摘胚乳发育期处于细胞形期的材料。在无菌条件下解剖种子,分离胚和胚乳。把胚乳单独接种于 B_5基本培养基附加 BA(2ppm)及2,4-D(0.5ppm)等培养基上,置于白天加1000 lux 的人工光照下培养,诱导出愈伤组织。将愈伤组织转移到 MS基本培养基附加 Ze(0·25ppm)及 NAA(0·lppm)培养基上,分化茎或根。它们在含附加 Ze(2ppm)及 IAA(1ppm)的 MS 培养基中进一步形成完整小植株。


A STUDY ON FORMATION OF THE PLANTLETS IN ENDOSPERMS OF LOQUAT IN VITRO
Abstract:On the basis of observing interrelationship between the changes of exte- rior morphological character of loquat(Eriobotrya japonica Lindl.)young fruit with colour of the peels and their interior embryonic development processes,we easily sampled the materials,whose endosperm remained in the developmental stage of cellular shape,through reconizing exterior char- acter of young fruits.The endosperm callus has been induced from the endo- sperm,which was separated from embryo sac and only inoculated on the B_5 basal media supplemented with BA(2ppm)and 2,4-D(0.5ppm)with duration of about 1000 lux light for daytime.When the callus was transfer- red to the MS basal media added to NAA(0.1 ppm)and Ze(0.25 ppm), the differentiation of shoots or roots began to occur.They further developed to perfect plantlets under the condition of employing MS addition with higher level of cytokinin.
