茶园土壤根际抗酸铝真菌A1F-1总RNA的提取

摘    要:用总RNA提取试剂盒提取经Al2(SO4)3100mol/L诱导和未经诱导的抗酸铝真菌ALF-1的总RNA,结果表明,预培养时间超过10小时,总RNA含量下降;在冰浴条件下提取RNA质量优于常温条件下提取的RNA;为了获得优质总RNA,ALF-1菌的预总培养和诱导时间分别以10h和5h为宜,整个RNA提取过程必须在冰浴条件下进行。

Abstract:Total RNA was extracted with total RNA extraction kit from Al induced and non induced fungus ALF 1, an Al resistant fungus separated from tea rhizospheric soil. The results showed that the total RNA concentration decreased when pre culture time exceeded 10h. The quality total RNA extracted under ice bath condition was higher than that extracted under room temperature.In order to obtain quality total RNA, the pre culture time and Al inducing time should be 10h and 5h respectively, and the extraction be on ice bath.
