荧光原位PCR定位动物精子介导转移的外源基因

摘    要:用荧光原位PCR对经体外介导外源基因转移方法处理的家畜精子进行检测。结果证实,牛,山羊和绵羊的精子在一定条件下能够携带所转移的外源基因,其平均转移效率分别为:绵羊27.12%,牛31.16%,山羊30.57%,同时发现外源基因已进入精子细胞内并位于核及其外围区域。

Abstract:In order to located the exogenous DNA molecule intosperm cells,we used fluo rescent in situ PCR technique to determinate the efficiency of sperms of the livestock as vector mediated the exogenous DNA transfer into sperm cells.The results showed that the sperm cells are able to take up exogenous DNA molecule in cattle,sheep and goat at experiment conditions,internalize into sperm''s nuclei,and locateat its per ipheral to the nucleus region.The average efficiency for DNA binding is 27.12% in sheep,31.16% in cattle and 30.57% in goat.
