淀粉凝胶电泳法对桑过氧化物同工酶研究

摘    要:用淀粉凝胶作支持物的平板电泳法,以叶、芽和茎皮为材料作桑若干品种的过氧化物同工酶分析。结果,其同工酶谱在同一品种不同器官间表现出不同,而同一品种同一器官在不同植株间则无差异;用作分析的材料以芽最好,经本方法同工酶分析鉴定,R81—1和R81—2是以新一之漱为亲本的遗传性变异的突变体。在桑同工酶分析研究上,用淀粉凝胶作支持物的平板电泳法比以往采用的聚丙烯酰胺法要好,不仅表现在效果好、成本低,而且在正负两极的凝胶板上均能显现出酶带,一次电泳还能分析好几种同工酶。

Abstract:Peroxidase isoenzymes of leaf, bud and shoot bark are determined for some mulberry varieties by the means of the tabular electrophoresis with the supporter of starch gel. The results are that the peroxidase isoenzyme patterns show the difference in different organs of same variety, no difference in same organ of different plants of same variety, and the best material for the analysis is the bud. R81-1 and R81-2 are the mutants of genetic variation from the parent Shinichinose through the experiment identification. The method with the starch gel is better than the method with the polyac-rylamide gcl for mulberry isoenzyme analysis, which not only showes the better effect and the lower cost but demonstrates isoenzyme bands on the both of positive and negative electrodes, and it can be used to analyses a number of isoenzymes through a same electrophoresis.
