中药连翘对大肠杆菌R质粒消除作用的研究

目的：探讨中药连翘对大肠杆菌R质粒的消除作用,使其重新恢复对抗生素的敏感性。方法：将大肠杆菌DH5α制备成感受态后,导入含有Ampr基因的质粒,再用连翘作用转化后的大肠杆菌,经培养12h后,挑出亚抑菌浓度的菌落涂于含有Amp的平板上培养24h,观察有无细菌生长,并通过电泳检测是否消除质粒。结果：在含有Amp平板上无大肠杆菌生长,在不含Amp的平板上细菌生长良好,质粒DNA电泳检测结果经中药作用过的大肠杆菌无质粒带,表明连翘对质粒具有消除作用。
