肉牛CAST基因分子多态性与肉品质性状相关性的研究

摘    要:运用现代分子生物技术寻找影响牛肉嫩度和大理石纹性状的候选基因。选取7个牛品种(群体)共176头牛作为试验动物,利用PCR—RFLP方法研究钙激活蛋白酶抑制蛋白基因(calpastatin,CAST)第6内含子(intron6)内的遗传变异,并进行克隆测序,证实酶切位点突变。利用最小二乘线性模型进行基因性效应差异显著性检验,结果表明CAST基因对牛肉的嫩度有显著作用,这为CAST作为牛肉嫩度候选基因提供进一步根据。

Abstract:This study was aimed to detect the genentic variations of CAST gene intron6 by PCR-RFLP in a total of 176 cattle from 7 breeds.After the polymorphism fragments were cloned and sequenced,the single nucleotide position and genotypes were assigned as BB for which being with the restriction site in CAST.Analysis of variance was conducted and it showed that beef tenderness was significantly influenced by CAST gene.
