小麦T型细胞质雄性不育恢复基因Rf6的ISSR标记分析

摘    要: 以小麦T型恢复系 2 114和不育系ND4 4A配制杂交组合 ,并以 14 7株个体组成的F2 分离群体作为恢复基因的标记群体。用 4 3个ISSR引物对两个亲本进行了扩增 ,发现 18个引物能在两者间产生明显的多态性 ,其中引物UBC 80 8、UBC 84 8在 2个亲本间以及可育池和不育池间都能产生一致、稳定的多态性。用这 2个引物在F2群体中进行扩增。经连锁分析 ,证明这 2个标记与小麦T型细胞质雄性不育恢复基因Rf6连锁 ,遗传距离分别为7.9cM和 4 .9cM。用 181对SSR引物对两个亲本进行扫描 ,其中有 34.3%SSR引物能在亲本间扩增出多态性 ,但没找到与Rf6连锁的SSR标记。对ISSR、SSR等方法在小麦中的多态性比例进行了比较 ,发现ISSR具有更高的多态性 ,特别是在外源基因的检测中能提供更丰富的遗传信息。

Abstract:Inter simple sequence repeat (ISSR) was evaluated in a F 2 population of 147 plants derived from male fertility restorer line 2114 and male sterile line ND44A. Of 43 examined primers, 18 produced distinguishable as well as polymorphic bands between the two parents. Linkage analysis in the mapping population showed that two markers, UBC 808 and UBC 848, were closely linked with the restoring gene Rf6 of the T.timopheevii CMS system. The distance between the two markers and the restoring gene was 7.9 cM and 4.9 cM, respectively. Two parental plants were screened with 181 pairs of SSR primers, of which, 34.3% showed polymorphism. No locus was found linked with the restoring gene. Compared with SSR technique that used in the experiment, ISSR produced more information and proved to be a valuable method for identification of alien fragment.
