栗不同品种过氧化物酶同工酶研究

摘    要:本试验采用聚丙烯酰胺凝胶电泳法,检测了板栗和日本栗两个种的41个品种(系)叶片的过氧化物酶同工酶,结果表明,栗品种叶片的过氧化物同工酶可分为二大酶区,第Ⅰ酶区相对泳动率(Rf)范围为0.056-0.272,共检出6条酶带;第Ⅱ酶区Rf=0.478-0.647,共检出8条酶带。根据各覆盖的出现频率,提高出了栗品种叶片过氧化物酶同工酶的基本酶谱,并采用信息量凝聚聚类法,初步绘出了栗品种间的亲缘关系图,认为我国板栗与日本栗间的亲缘关系比较接近。

Abstract:Isoperoxidase of mature leaf of 41 varieties or strains from Chinese Chestnut and Japanese Chestnut was examined by polyacrylamide gel plate electrophoresis.The result indicated that the isoperoxidase of the varieties or strains in chestnut could be divided into 2 regions. In the first region of slow mobility, the zone of Rf value was from 0.056 to 0.272.Six isoperoxidase bands were distributed within this region.In the second region of fast mobility, Rf value was from 0.478 to 0.647.Eight isoperoxidase bands were distributed within this region.According to the frequency of each isozyme band, the basic zymogram of isoperoxidase of the varieties in chestnut was suggested. A picture of the relationship between the varieties or strains in chestnut was made by information content hierarchical agglomeration clustering.The relationship between Chinese Chestnut and Japanese Chestnut was considered closer.
