HBV RNA复制子疫苗、DNA疫苗在BHK-21细胞中表达情况的比较

摘    要:为建立更有效的乙肝疫苗,通过将所构建的HBVRNA疫苗和DNA疫苗分别转染BHK-21细胞,并比较其在细胞中表达情况及细胞凋亡现象。以pSFV为基础构建的疫苗载体的表达效率高于以pcDNA3.1为基础构建的疫苗载体,pSFV为基础构建的疫苗载体出现细胞凋亡的现象。HBVRNA疫苗比DNA疫苗表达效果更好,安全性更高,更具有应用前景。

Abstract:Abstract: Construction of RNA replicon vaccine and DNA vaccine of HBV, then the 2 vectors were tranfected into BHK-21 cells and their expression efficiency was detected by ELISA with the kits used to detect HBsAg and preS2. Meantime, we found that the vectors based on pSFV can make BHK-21 cells dead (apoptosis). It is confirmed that the immune effect of RNA replicon vaccine is better than that of DNA vaccine. RNA vaccine is safer and more prospects.
