摘    要:目的]为建立一种快速检测动物血液中SAL残留的荧光光度检测法。方法]以猪血清为样品,化学法进行前处理,用荧光光度法选择SAL的最佳提取溶剂,研究样品pH值、提取溶剂用量对提取率的影响。结果]SAL的最佳提取溶剂是异丁醇。样品的最佳pH值为10.00。随着异丁醇用量的增加,SAL提取率不断提高,当异丁醇用量超过60ml时,提取率几乎不再变化。荧光光度法的检出限是0.97ng/ml,异丁醇的线性范围是0~12.8μg/ml,相对标准偏差为5.2%,样品的加标回收率为98.0%~104.2%。结论]荧光光度法样品前处理简单,选择性强,用样量少,操作方便,灵敏度高,整个过程只需15~20min,线性范围较宽,精密度和回收率良好,适合现场检测。

Abstract:Objective] The research aimed to set up a kind of fluorescence detection method for detecting SAL residue in animal blood rapidly.Method] With porcine serum as samples,pretreatment was carried out by chemical method and the optimum extraction solvent for SAL was selected by fluorophotometry.And the effects of pH value of samples and the dosage of extraction solvent on the extraction rate were studied.Result] The optimum extraction solvent for SAL was isobutanol.The optimum pH value of samples was 10.The extraction rate of SAL was continuously increased with the increment of isobutanol dosage and the extraction rate hardly changed when the dosage of isobutanol exceeded 60 ml.The detection limit of fluorophotometry was 0.97 ng/ml,the linear range of isobutanol was 0~12.8 μg/ml,the relative standard deviation was 5.2% and the recovery rate of sample with adding standard substance was 98.0 %~104.2 %.Conclusion] The fluorophotometry method has such features as simple sample pretreatment,strong selectivity,little sample,simple operation and high sensitivity.The total process of this method only needs 15~20 min and it had wide linear range and good precision and recovery rate,so it is suitable for on-site detection.
