Abstract:Cymbidium mosaic virus (CymMV) is one of the most important viruses that cause orchids great losses. Triple gene block protein 1 (TGBp1) is a multifunctional protein of CymMV. Homologous interaction of CymMV TGBp1 is essential for its biological functions, but little is known about the site of homologous interaction of TGBp1. In this study, three truncated mutants of TGBp1-1 (motif I, Ia), TGBp1-2 (motif II, III, IV) and TGBp1-3 (motif V, VI) were constructed based on its motifs respectively. Yeast two-hybrid (Y2H), bimolecular fluorescence complementation (BiFC) and GST pull-down were used to screen the site of homologous interaction among three truncated mutants of TGBp1 and TGBp1. The results showed that TGBp1-2 (motif II, III, IV) was the main site of homologous interaction of TGBp1. Our results provide a foundation for further research on the movement and replication of CymMV.
