卷叶贝母愈伤组织超低温保存关键技术研究

摘    要:[目的]建立一套完整、快速和经济的卷叶贝母超低温保存关键技术。[方法]对卷叶贝母愈伤组织超低温保存技术中的预培养时间、保护剂类型和解冻方式进行了研究。[结果]预培养时间、冷冻保护剂类型和解冻方式均是影响卷叶贝母愈伤组织冷冻保存过程中的关键因素。经预培养9 d、由10%二甲基亚砜和0.5 mol/L山梨醇组合的复合冷冻保护剂冷冻、在40℃水浴条件下解冻的卷叶贝母愈伤组织其保存效果最佳,保存后细胞的存活率可达87.39%。[结论]为更好地保护卷叶贝母种质资源提供了参考。

The Key Cryopreservation Techniques of Fritillaria cirrhosa D.Don Callus

Abstract:[Objective] To establish an integral,rapid and economic key technique for the cryopreservation of Fritillaria cirrhosa D.Don callus.[Method] The pre-culture time,cryopreservation medium and unfreezing method of the key cryoperservation techniques of Fritillaria cirrhosa D.Don callus were studied.[Result] The pre-culture time,cryopreservation medium and unfreezing method were the key factors during the cryopreservation of Fritillaria cirrhosa D.Don callus.The optimal preservation conditions were pre-culturing for 9 days,10% DMSO containing 0.5 mol/L sorbitol as the cryopreservation medium,and unfreezing at 40 ℃ water bath.The survival rate of the cells of Fritillaria cirrhosa D.Don callus preserved by this way could reach to 87.39%.[Conclusion] The study provided reference for beter preserring the resources of Fritillaria cirrhosa D.Don.
