拟鲶高原鳅黄河和大通河种群的遗传多样性和分化水平研究

摘    要:目的]探究拟鲶高原鳅(Triplophysa siluroides)黄河种群与大通河种群的遗传多样性和分化水平。方法]采用聚合酶链式反应(PCR)和直接测序方法获得拟鲶高原鳅线粒体DNA(mtDNA)细胞色素b(Cytb)基因的全序列。结果]拟鲶高原鳅2个种群共47个个体均获得Cytb基因的全长序列(1140 bp),共定义了12个单倍型。拟鲶高原鳅黄河种群遗传多样性低于大通河种群。结论]人类活动的影响和引入外来种可能引起了拟鲶高原鳅黄河种群遗传多样性的下降;同时,拟鲶高原鳅黄河种群和大通河种群间存在显著的分化,建议在实践中应将2种群分开管理和保护。

Distribution and Properties of Amylase in Alimentary Tract and Hepatopancreas of T.siluroides

Abstract:Objective] The research aimed to investigate the genetic diversity and differentiation of T.siluroides populations from Yellow River and Daitong River.Method] The complete sequences of the mitochondrial cytochrome b gene of T.siluroides were obtained by using of PCR and DNA sequencing.Result] The complete sequences of Cyt b gene(1 140 bp) were obtained from forty seven individuals of T.siluroides,representing two populations from Yellow River and Daitong River,and a total of 12 haplotypes were identified.The genetic diversity of population from Yellow River was low than that of population from Daitong River.Conclusion] The decline in genetic diversity of T.siluroides population maybe related to human activities and introduction of exotic fishes.The significant genetic differentiation between populations from Yellow River and Daitong River suggested that the two populations should be managed and preserved separately.
