摘    要:目的]对1株抑烟草青枯病生防菌进行筛选、鉴定和活性物质研究。方法]采用系列稀释法和平板涂布法从黔江烟区土壤中分离得到菌株,并通过薄层层析、硅胶柱层析、HPLC等手段对其活性物质进行研究。结果]分离得到1株编号为5B18的菌株,其对烟草青枯病病原菌(Ralstonia Solancearum)有强烈拮抗作用。全细胞脂肪酸及16SrDNA分析结果表明,该菌为解淀粉芽孢杆菌(Bacillus amyloliquefaciens)。对该菌提取物分析表明,该提取物为13种不同的化合物组成的混合物。结论]该研究为从微生物角度防治烟草青枯病提供理论基础。

Study on the Selection,Identification and Its Active Substances of A Biocontroi Bacterium against Tobacco Bacterial Wilt

Abstract:Objective]The paper was to study the selection,identification and its active substances of a biocontrol bacterium against tobacco bacterial wilt.Method]The strain was isolated from the soil of tobacco cultivation region in Qianjiang by using serial dilution method and spread-plate method.The strain's active substances were studied by TLC,silica gel column chromatography and HPLC.Result]A strain marked as 5B18 was isolated and proved to have significant antagonism to Ralstonia Solancearum.The whole cell fatty acid and 16SrDNA analysis indicated the strain was Bacillus amyloliquefaciens.Analysis of the strain extract showed that the extract was a complicated mixture of 13 different compounds.Conclusion]The research could provide theoretical foundation for the control of tobacco bacterial wilt from the perspective of microbiology.
