旱稻原生质体培养和植株再生的研究

摘    要:旱稻仿野生87-707品系具有多年生趋势,是我院研究多年生旱稻的一个材料。我们用它进行原生质体培养,想为研究多年生旱稻基因改良打下基础。现用该品系的成熟胚诱导的愈伤组织,在含有氨基酸丰富的AA培养基中进行细胞悬浮培养。取该细胞悬浮培养物游离的原生质体,用琼脂糖包埋于改良的KM8P培养基中,发生了持续分裂。经一系

Abstract:Fang Ye 87-707 species of upland rice has the potential of perennial character, and thus serves as a material of the perennial upland rice in our Academy. We use it for protoplast culture and hope it lay a basis for studying genetic improvement of perennial upland rice. By using the callus induction of mature embryo to conduct suspension cell culture in the AA medium with abundant amino acid, the isolated protoplast in the suspension cell culture was embedded with a-garose in improved KM8P medium, the sustained division is occured. By a serial of alternation into differentiation medium, the protoplast underwent into a complete plant and was successful in transplanting. We have made some improvements on isolation and culture method as follows: 1) Adding ABA into the suspension cell medium to benefit the survival and division of the protoplast. 2) It is of great importance for the success of isolation to find the best condition of suspension cell before the isolation of protoplast. 3) The agarose embedding culture greatly increases the plating efficiency. 4) By using higher osmotic pressure of medium to culture regenerated cell colony and callus of the protoplast is beneficial to raise the regeneration ability of plant. 5) By alternating differentiation medium will significantly raise he regenerated frequency of plant. This is so far the first report about the regenerated plant from upland rice protoplast both at home and abroad.
