盾叶薯蓣愈伤组织诱导研究

摘    要:以试管培养的盾叶薯蓣为材料,研究了基因型、外植体种类、光暗培养条件、激素配比等因子对其愈伤组织诱导的影响。结果表明:不同基因型盾叶薯蓣的愈伤组织诱导率有较大差异;同一基因型,4种外植体(微块茎、幼芽、叶片、胚)的愈伤组织诱导率有所不同,胚和微块茎易诱导出愈伤,幼芽居中,而叶片较难,光暗培养条件下不同外植体的愈伤组织诱导率有差异,暗培养对叶片愈伤诱导有利,而光培养有利于微块茎愈伤组织的形成;2,4-D可以诱导微块茎、幼芽、胚形成愈伤,但6-BA和2,4-D的组合可以显著提高愈伤组织诱导率。叶片愈伤组织的诱导需要添加NAA和6-BA。

Abstract:The effects of genotypes, explants, phytohormone combination and light condition on callus induction of D. zingibernsis were studied.Different genotypes of D. zingibernsis displayed diversity in callus inducement. Four types of explants also displayed various abilities in callus formation. Embryo and shoot-tip exhibited high callus inducing percentage, followed by mircrotubes while leaves were difficult to induce callus. Light culture condition provided different influence on callus inducement to different explants. Dark condition enhanced callus formation in leaf segments but this condition showed negative effect on callus formation in microtubes, which were enhanced in light cultured condition. To different explants, phytohormone combinations should be modified to get more effective callus inducement: increasing of concentration of 2,4-D to some extent could promote callus inducement in most of explants, but it was inefficient to callus inducing in leaf segments, which need addition of NAA for callus formation. The combination of 6-BA and auxin could remarkably enhance the callus inducement.
