摘    要:联合使用截留相对分子质量1000的超滤膜和270的纳滤膜从刺芹侧耳(Pleurotus eryngii)水提液中分离海藻糖。超滤膜操作压力为4 kg/cm2,同时纳滤膜操作压力为3 kg/cm2,经膜分离的相对分子质量270~1000部分溶液经浓缩结晶,海藻糖纯度可达93.3%。

Separation of Trehalose from Aqueous Extracts of Pleurotus eryngii Fruit Bodies Using Membrane Filtration
Abstract:Pleurotuseryngii fruit bodies (1 kg)were extracted twice with ddH2O at 100 ℃ for 2 h (fruit body∶water ratio was 1∶15 w/v for the first extraction and 1∶10 w/v for the second).Combined extracts (total volume~18 L)were filtered through cheese cloth and the filtrate was separated into three fractions using a polyether sulphone membrane filter (PES1)(MWCO 1000 Da)and a NF270 nanofilter (MWCO 270 Da). Pressures of 4 kg/cm2 and 3 kg/cm2 were applied when filtering through the PESI and NF270 membranes, respectively.The fraction passing through the PESI membrane but retained by the NF270 nanofilter was concentrated using a rotary evaporator and the trehalose concentration in this fraction was adjusted to 30%. Five volumes (v/v)of absolute ethanol were added and,after mixing,the solution was left to stand overnight at 4 ℃.Precipitated material was collected by suction filtration and re-crystallized from water∶absolute ethanol (1∶5,v/v)to give a trehalose sample of 93.3% purity.
