摘    要:为探究ABC转运蛋白介导毛白杨叶际异常维克汉姆酵母对BDE-3的耐受机制,通过对其中Wapdr15基因克隆、基因序列及编码蛋白质结构分析、烟草遗传转化以及转Wapdr15基因烟草的BDE-3暴露胁迫实验,发现Wapdr15基因编码序列长4536 bp,编码蛋白具有TMD1-NBD1-TMD2-NBD 2结构,为全分子跨膜蛋白;转Wapdr15基因烟草组培苗叶片对BDE-3的耐受性显著提高,推测Wapdr15参与了毛白杨叶际异常维克汉姆酵母对BDE-3的解毒耐受过程。

Abstract:In order to explore whether the ABC transporter mediates the tolerance mechanism of Wickerhamomyces anomalus isolated from Populus tomentosa phyllosphere to BDE-3 exposure,the Wapdr15 gene was cloned,sequenced and analyzed on its coding protein structure.The results showed that the length of the coding sequence of Wapdr15 gene was 4536bp,encoding a full molecular transmembrane protein with the structure of TMD1-NBD1-TMD2-NBD2.Tobacco tissue culture seedlings with Wapdr15 gene were obtained via agrobacterium-mediated transformation,and the BDE-3 exposure stress test showed a significant increase in tolerance to BDE-3 compared to the control,revealing that the Wapdr15 might be involved in the process of detoxification of W.anomalus to BDE-3.
