摘    要:根据水稻腥黑粉菌两个聚类群与其它黑粉菌ITS序列的差异,分别设计了两个聚类群的特异引物Hor2/Hor9和Hm1/Hm5,结合ITS通用引物Til1/Til4建立了分别检测水稻腥黑粉菌两个聚类群单个冬孢子的套式(巢式)PCR检测方法,整个检测过程可以缩短至8h。

Abstract:Isolates of Tilletia horrida from different locations were divided into two distinct clades(groups) based on the rDNA ITS sequences,with the sequence difference,the nested PCR methods using group-specific primers Til1/Til4(Hor2/Hor9) and Til1/Til4(Hm1/Hm5) were developed for detecting teliospore of two groups, respectively. One teliospore can be amplified and obtained specific amplicon. The process of detection can be finished within 8 hours. Because of avoidance of DNA extraction,this method can be useful and potential for detection of a few teliospores in imported grain samples.
