利用PCR技术检测鸡肉产品中的空肠弯曲杆菌

摘    要:根据空肠弯曲杆菌旋转酶基因(gyrAgene)的保守序列,设计1对引物,建立了快速检测鸡肉中空肠弯曲杆菌的PCR方法。采用该方法和生化试验对送检的100份鸡肉样品进行检测,结果表明:PCR方法能特异性地扩增出空肠弯曲杆菌192bp核苷酸片段,其它结肠弯曲杆菌、肠炎沙门氏菌、大肠杆菌、副溶血性弧菌、产气荚膜梭菌、绿脓假单胞菌等均不能获得阳性扩增结果。检测限度为8cfu/mL。鸡肉肉水、增菌液和纯培养菌落的PCR鉴定结果,阳性率分别为16%、24%和20%,生化鉴定检出率为20%。

Abstract:A rapid polymerase chain reaction(PCR) method was established to detect C.jejuni in chicken meat with a pair of primers designed and synthesized according to the sequence of C.jejuni gyrA gene.100 chicken meat samples were detected with the method and API Campy kit.The result showed that 192 bp was specifically amplified from template of C.jejuni,but did not amplify from those of C.coli,S.enteritidis,E.coli,V.parahaemolyticus,C.perfringens,A.Bacillus,and so on.The limit of the method to detect the bacteria was 8 bacteria cfu/mL.Chicken meat water,enrichment fluid and colony from the pure culture were detected by PCR,and 16%,24%,20% of them were found respectively to be positive for C.jejuni.The detection rate of biochemistry identification is 20%.
