脱硫菌胞外聚合物提取及分泌量影响因素研究

摘    要:[目的]选择一种较优的黄单胞杆菌EPS提取方法,并分析环境因素对EPS分泌量的影响。[方法]分别采用离心、加热、超声、硫酸、EDTA和甲醛加碱法提取EPS,测定提取物中多糖、蛋白质及DNA含量,以评价提取效率及对细胞的破坏程度。利用正交试验设计考察温度、培养基pH值等因素对EPS分泌量的影响。[结果]甲醛加碱法和超声法的提取效率较高。通过对不同超声功率的比较,选择在50W下超声4min提取EPS。正交试验结果表明,黄单胞杆菌EPS最大分泌量的环境条件为pH值6.0,温度32℃,菌液比7∶100,金属盐溶液投加量3ml。[结论]黄单胞杆菌EPS提取采用超声法;环境因素中温度对分泌量的影响最大。
