摘    要:以吉林地区三种金针菇以及河南产地的金针菇为材料,测定、分析了金针菇rDNA-ITS序列,并通过与GenBank上已登录的金针菇ITS序列比对,采用邻接法(Neighbor Joining,NJ)构建系统发育树。结果表明,所有供试材料ITS全长在712 bp~719 bp范围内,G+C含量在49.4%-49.9%之间,各样品间有35个变异位点,13个信息位点,遗传距离在0~0.014之间,其中吉林地区3种金针菇样品部分位点发生突变,均没有同步进化。9个金针菇样品的ITS序列被分成了两大组,其余样品之间又出现很多分支,表明ITS可作为金针菇亲缘关系鉴定的分子标记。

Abstract:The rDNA-ITS sequences of there kinds of Flammulina velutiper in Jilin area and Henan origin Flammulina velutiper were measured, analysed. Contrasted the sequence of Flammulina velutiper having been logged in GenBank and adopted the neighbor-joining method to construct the phylogenetic tree. The results show that the total length of all the tested material internal transcribed spacer is within the scope of 712 bp-719 bp, the content of G+C is between 49.4% ~ 49.9%, there are 35 variable sites and 13 informative sites among all sorts of samples, the range of genetic distances is between 0~0.014, thereinto, apart of the there kinds of Flammulina velutiper sites in Jilin area appeared to be mutating, and none of them evolved synchronous. The Nine kinds of Flammulina velutipes could be obviously divided into two branches, other samples took on many branches. This show that internal transcribed spacer can be the molecular sign to authenticate the phylogenetic relationship of Flammulina velutiper.
