PM2.5对大鼠血管平滑肌细胞PCNA、VCAM-1表达及增殖的影响

摘    要:目的观察大气细颗粒物(PM2.5)对培养的大鼠血管平滑肌细胞(VSMC)增殖细胞核抗原(PCNA)、血管细胞黏附分子-1(VCAM-1)表达及增殖的影响,了解其可能的作用机制。方法采集湛江市区大气中的PM2.5,以10、20、50、100、200mg/L剂量染毒培养的大鼠VSMC24h后,观察PM2.5对VSMC的PCNA、VCAM-1表达(免疫组化法)及增殖(MTT法)影响。结果在10-200mg/L范围内,随着染毒质量浓度增加,VSMC的PCNA水平从(16.67±2.58)上升到(58.00±8.25)(F=82.23,P〈0.01),VCAM-1水平从(15.83±2.32)上升到(60.83±2.56)(F=258.63,P〈0.01),MTT值从(0.49±0.067)增加到(0.72±0.025)(F=27.98,P〈0.01)。结论PM2.5促进体外培养的VSMCPCNA、VCAM-1表达,刺激VSMC增殖,在10-200mg/L质量浓度范围内具有剂量依赖性。
