秋水仙素对刺葡萄的诱变

摘    要:以刺葡萄(Vitis dabidiiFoex)为试材,利用不同浓度的秋水仙素溶液,采取浸润法与注射法处理生长芽,观察其叶片与枝蔓的变化,测定其气孔、花粉与叶绿素,并进行染色体检测。结果表明:秋水仙素对刺葡萄诱变效果良好,其中以0.3%秋水仙素处理获得最佳效果,变异率为33.3%;染色体加倍的变异枝条叶片增厚26.7%-32.0%,气孔密度减少,保卫细胞增大,花粉粒增大38.4%,节间变短,叶片叶绿素含量显著增加;染色体出现2n=4x=76及非整倍体现象。

Abstract:Taking the diploid Vitis dabidii as materials,using different concentrations of colchicines,with immersion of colchicine concentration on axillary bud and injection of colchicine concentration on axillary bud,variation in leaves and branches was observed,leaves stoma,pollen,chlorophyll were measured and chromosome of V.dabidii were tested.The results showed that the mutagenic effects of colchicines on V.davidii were remarkable,0.3% colchicines brought the most significant effects,its respective inducement ratio was 33.3%.The mutated branches had thicker leaf(26.7%-32.0%),the stomata density of the leaf had become low,the size of defensive cell increased,and the pollen size increased by 38.4%.The branch segment became shorter.There has been an appreciable increase in chlorophyll,the chorosome number of mutated planllets was 2n=4x=76,and some mixoploids were obtained.
