摘    要:以空间诱变水稻H4成熟胚为外植体,通过调节2,4-D浓度、激素组合和分化途径,建立适宜的再生体系.结果表明:诱导培养基以NMB为基本培养基,添加2.5 mg/L 2,4-D、1.0 mg/L NAA和0.5 mg/L 6-BA,26℃暗培养30 d,愈伤诱导率达91.63%;继代培养基中添加2 mg/L ABA,对胚性愈伤组织的形成及后续分化率有一定的促进作用;两步分化法可提高愈伤组织的分化效率,分化率达84.76%,植株再生率为77.67%.初步建立了H4成熟胚愈伤组织的再生体系,为后续的遗传转化研究奠定基础.

Abstract:To establish a suitable regeneration system of space-induced indica rice, H4, a series of experimental programs were carried out. The results showed that When NMB was used as the basic medium, 2.5 mg/L 2,4-D, 1.0 mg/L NAA and 0.5 mg/L 6-BA were added, the callus induction frequency of H4 was 91.63%; ABA showed a significant result on the growth and regeneration of callus; the two -step culture method exhibited positive effects on enhancing plant regeneration ability. The regeneration technology for high frequency of H4 mature embryos was established preliminarily.
