桑树花药愈伤组织诱导方法初步研究

目的]对桑树花药愈伤组织诱导方法进行研究.方法]以MS为基本培养基,研究不同品种、不同预处理时间和不同的激素种类及浓度对愈伤组织诱导的影响.结果]不同品种适于花药培养的花序大小有所差异,同品种内长度相对稳定,可以作为取材标准;低温预处理对试验的结果影响不显著（P＜0.05）;不同品种间诱导率差异较大,在供试的3个品种中,桂桑优62的诱导率最高,为46.4％,最低的为粤桑11,仅为28.8％;激素对愈伤组织诱导率影响较大,特别是2,4-D,最佳激素组合为1.0 mg/L2,4-D ＋0.5 mg/L 6-BA.结论]该方法研究了桑树花药愈伤组织的诱导方法,为桑树的进一步开发研究提供了依据.

Preliminary Study on Callus Inducement of Anther Culture in Mulberry

Objective ] To study callus inducement of anther culture in mulberry. Methods ] Using MS as basic medium with exogenous hormone to study anther callus inducement and discuss influences of different varieties, pre-treatment time and hormone. Results] The length of inflorescence suitable for culture was significantly different among the varieties, however, in the same variety, the length was a stable character. The influence of pre-treatment at a low temperature was not significant. The callus induction rate was different among the three varieties, the variety ＂Guisangyou 62＂ with the highest 46.4% and ＂Yuesang 1 1＂ with the lowest 28.8%. The combination and concentration of hormones were important factors, especially 2,4-D, and the best group is 1.0 mg,./L 2,4-D ＋ 0.5 mg/L 6-BA. Conclusion ] This study discussed callus inducement of anther culture in mulberry, to provide basis for further development and study of mulberry.
