滇独蒜兰的组织培养研究*

摘    要: 以滇独蒜兰的不同部位为外植体进行组织培养方法的研究,结果表明:以种子为外植体最佳,种子在MS+NAA0.2 mg/L 的培养基在上经过2~3个月的培养,有一部分的种子开始形成极小的愈伤组织或原球茎;再转接到MS+BA5.0~10.0 mg/L+NAA0.2 mg/L 的培养基上,其中少部分会进一步分化成胚状体,并形成小植株,经过多次继代转接可形成丛芽,丛芽的增殖系数高达6倍。通过进一步优化培养基及培养条件,可使种子培养的效率提高。以叶片和鳞茎为外植体,均未获得组培苗。

Abstract:The diffferent materials of Pleione yunnanensis are used as explants for tissue culture. The result shows the seeds are the best materials. Some seeds begin to produce very small callus or green globlar body while the seeds at the medium of MS+NAA0.2 mg/L after two-three months. A few of them can be induced to form embryoids, and then form the young plantlets when they are transformed to medium of MS+BA 5.0~10.0 mg/L+NAA0.2 mg/L . There are shoots formed after many times subculture in the same medium. Multiplication rate was near six times. The effect of seeds tissue culture can be improved by optimuming of the medium and the condition. When leafs and bulbs are used as explants the experiment is fail.
