铜缺乏奶牛肝脏金属硫蛋白的层析特征

摘    要:为阐明金属硫蛋白在铜缺乏奶牛肝脏中的代谢特征,选择铜缺乏区有明显临床症状的奶牛6头,采集肝脏样品后用差速离心法分离肝脏金属硫蛋白,以Sephadex-G75进行层析纯化,测定洗脱液中蛋白质、巯基含量、紫外光扫描值、216nm和260nm处吸收峰值及荧光扫描值。将6头健康奶牛的肝脏样品作为对照。结果表明,发病奶牛肝细胞浆Sephadex-G75凝胶层析液第1峰值各管中蛋白含量显著高于对照组,但第2、第3峰值各管中蛋白含量显著低于对照组;巯基含量的检测也显示了类似的结果。发病奶牛紫外吸收值及荧光吸收值出现第2峰或第3峰,且其含量显著高于对照。结论:铜缺乏能够改变奶牛肝脏中金属硫蛋白的代谢。

Abstract:6 cows with evident syndrome of sway back from the areas of copper deficiency were selected and metallothionein was isolated from hepatic sample by differential centrifugation and purified by Sephadex-G75 gel column chromatography to clarify the metabolic features of metallothionein in liver of cows with copper deficiency. The concentrations of protein,-SH and absorption value of A216,A260 and flourence in the fraction of Sephadex-G75 chromatography were determined while the specimens of liver were collected as control from 6 clinically healthy cows. The results showed that the contents of protein in the fraction of the first peak were significantly lower than that of control group while those in second and third one were more higher in comparision with that of control group. Similar results were found while determining the content of the -SH. The absorption features of A216,A260 and flourence in the fraction from morbid cows were manifested by the obvious appearance of the second and third peak and the value of the two peaks was higher than that from healthy ones, conclusion : copper deficiency could change the metabolism of MT in the liver of cows with copper deficiency.
