兔动脉粥样硬化斑块与肾素-血管紧张素系统关系的研究

摘    要:为探讨肾素 -血管紧张素系统 ( RAS)对实验性兔动脉粥样硬化 ( AS)的影响 ,采用 2 0只雄性新西兰大白兔 ,随机分为 2组。对照组饲以全价颗粒饲料 ,实验组采用高胆固醇饮食( HCD)饲喂法建立 AS模型。分别检测总胆固醇( TC)、血管紧张素转换酶 ( ACE)和血管紧张素 ( Ang ) ,并对主动脉 AS病灶进行病理检查。结果表明 :与对照组相比 ,高胆固醇饮食 1周后 TC开始显著升高 ,第 4周达高峰 ,第 1 6周组织中 Ang 水平显著升高 ( P <0 .0 5) ;病理检查见主动脉内膜满布淡黄色斑块 ,镜检由增厚的内膜、泡沫细胞和胆固醇结晶组成 ;对照组无异常。高胆固醇饮食 1 6周可建立兔 AS模型 ,RAS可影响 AS斑块的形成

Abstract:Objective:The aim of this study is to examine the effect of renin-angiotensin-system (RAS) on the pathological process of atherosclerosis in rabbits.Methods:20 male New Zealand rabbits were divided randomly into two groups: control group and high-cholesterol-diet (HCD) group.Rabbit AS model was established by feeding with high cholesterol diet. The serum level of total cholesterol (TC),Angiotensin-1 converting enzyme (ACE) and AngII concentrations were the serum and tissue of examined simultaneously. Then the atherosclerotic aortas were processed and analyzed for histopathological change.Results:The levels of TC begun to rise at 1 week after high cholesterol intake and reached the peak level at the end of 4th week, which was 20 times higher than the beginning; the tissue concentrations of AngII was significantly higher than that of control group (P<0.05) in 16 weeks. Macro-pathological observation: The intima of control group was smooth and normal, but that of HCD group was scattered with yellow-colored lipid streaks and plaques. The atherosclerotic plaque was composed of thickened intima, foam cells and cholesterol crystal accumulation.Conclusions:The rabbit model of atherosclerosis may be successfully established by high cholesterol diet intake. The progress of atherosclerosis can be affected by RAS.
